BIOINFORMATICS METHODS FOR THE CHARACTERIZATION OF MURINE ADIPOSE-TISSUE DERIVED STROMAL CELLS ALTERED BY CONTINUOUS ROTATION SUSPENSION

Abstract
Abstract - The liver is one of the most susceptible organs in the human body, it is subject to a constant stress and a consistent damage can be promoted. In severe cases of liver disease, once the preservative measures are ineffective, the only option for patients is the liver transplant. Methods for producing functional differentiated cells suitable for cell therapy must be implemented.For maximizing the differentiationcapabilityof murine adipose tissue derived stromal cells (ADSCs), we established a stress inducing method bythe continuous rotation/suspension of ADSCs.This treatmentenhanced the stemness ability of ADSCs, we named these modified cells: Rota-ADSCs. We used methods of bioinformatics for a comprehensive characterization of Rota-ADSCs, which permitted the analysis of large amount of gene expression information by means of DNA microarray analysis.Finally, we confirmed that the use of Rota-ADSCs allowed efficient differentiation towards mesodermal and endodermal cells like adipocytes, osteocytes, and hepatocytes.
