Production of Biopolymer from Locally Isolated Bacteria in Kurdistan Region-Erbil/Iraq

Abstract
The study was aimed explore possibility of isolate microorganisms which isolated from different sources to produce a biopolymer. Bioplastics are considered as environmentally friendly will complete biodegradable after disposal. The collection of 130 isolates bacteria from different samples from Erbil city and neighbouring regions during November 2014 to February 2015, and screening for poly 3-hydroxybutyrate (P3HB) production, only 20 bacterial isolates have the ability for the P3HB production. Accordant to quantity and quality,one of isolates was selected to further study.According to the result of polymer chain reaction (PCR) the G1 isolate identified as Citrobacterbraakii, isolate.The quantity of product depended on cell dry weight percentage was ranged from (3.939 - 98.34 %). Also, estimation of (P3HB) concentration by crotonic acid assay (standard curve) the concentration of P3HB was ranged from (0.03394- 0.00868) g/l.Nuclear magnetic resonance (NMR) spectrum (1HNMR) technique was performed for obtaining information about the number and arrangement of hydrogen atoms in P3HB by their chemical shift data. It can be concludedthatthere are several strains of bacteriaable to produce biopolymer. More of these isolates were found from swage water and soil mountain sources.
