Co-Supporting Lipoic Acid and Resveratrol in Magnesium (Lipresv@MDH, Trade Name Revifast Lipo) Dihydroxide with Improved Pharmacokinetic Profile: An In Silico Study

Abstract
Problem statement Recently, our group demonstrated the enhancement of bioavailability of resveratrol when supported in magnesium dihydroxide (Resv@MDH, trade name Revifast)1. Now, we have investigated the effects of co- supporting on magnesium dihydroxideresveratrolalong with a molecule of Biopharmaceutical Classification System (BCS) class I, thealfa-lipoic acid (ALA). Methods The new molecule (23% of resveratrol w/w, 23% of α-lipoic acid w/w, and 54 % of magnesium hydroxide w/w) was subjected toin vitro studies such as dissolution tests and sspectrometry. Moreover, simulations of pharmacokinetic processes were implemented based on a model previously described by our group2. Results Dissolution testsshoweda complex solubility profile of lipoic acid in gastric fluid simulation withacomplex kinetic profilewithrespect to pure ALA, indicating the magnesium complex formation during the dissolution process of α-lipoic acid. A spectrophotometric study confirmed the magnesium-ALAinteraction as demonstrated by the variation of the absorption peak ataconcentration similar to gastric dissolution test solution. Thesimulation of pharmacokinetic processes showed that the magnesium-ALA complex increases the time of absorption and the time of α-lipoic acid lived in the plasma compartment. Conclusion Increasing the duration of α-lipoic acid represents an opportunity to improve the biological effects of this nutraceutical compound, especially in formulations containing substances with physiological synergic effects such as resveratrol. The synergic action of α-lipoic acid and resveratrol obtained byRevifastLiporepresents a promising nutritional ingredient against dysfunctions such as metabolic syndrome often associated to pathologieslikepolycystic ovary syndrome (PCOS).
