The Analysis of Escherichia Colio157:H7 Bacteriophage Host Specificity from Surabaya, Indonesia

Abstract
Escherichia coliO157:H7is one of the pathogenic bacteria causing foodborne disease. The use of lytic bacteriophages can be a good solution to overcome the disease. This study aimedto analyzedE. coliO157:H7 bacteriophage host specificity from Surabaya, Indonesia. The sample used in this study was eight bacteriophages from prior study, and the technique used in identifyingE. coliO157:H7bacteriophage host specificity was Mass Spectrometry and NCBI Blast.The result obtained peptides hit by NLSVEEQYTAAAAK, KLLSEGQDVTMHAK, and VDIEDTLAR in the molecular weight range of 22,030 kDa, which had a score range of 62 to 100, with 11 % to 18 % sequence coverage on bacteriophage host specificity. With high percentage of97.97 % to 99.49 % homology identity with Escherichia phage EC101, Escherichia phage vb_EcoM_bov9_1, Escherichia phage vb_EcoM-CH2BS1, Escherichia phage ECO4, Phage NBEco003, Escherichia phage vb_EcoM_WL-3, according to NCBI Blast.The conclusion obtained from this study is that according to NCBI, E. coli O157:H7lytic bacteriophages from Surabaya, Indonesia had a significant percentage to other Escherichia phage. Therefore,It could be a new biocontrol agent for E. coli O157:H7, which contaminates food and causes foodborne diseases. Keywords- E. coli O157:H7, Escheria Phage,Foodborne Pathogen, Host Specificity.
