Is it Dangerous to Use Extract of Mare Placenta for Humans Blood Mononuclear Cells?
Abstract
Equine placenta extract products have unique biological effects. That is why these products are actively used in various fields of medicine. The aim of this work was to study the effect of products based on horse placenta extract on human peripheral blood mononuclear cells.
Materials and methods: The study was conducted at the Kazakh National Medical University named after S.D. Asfendiyarov and in the Institute of Fundamental and Clinical Medicine named after B. Atchabarov
Placental extract of the placenta was taken from healthy horses. The collection of mononuclear elements was taken from the peripheral venous blood of healthy volunteers. To assess the level of harmful effects of the elements of the placental extract, a colorimetric test was used. For control, we used cultures of mononuclear cells without contact with the investigated extract. The toxicity of the extract was studied using propidium iodide. The effects of reactive oxygen species were also evaluated. To determine the volume of secretion of interleukins, the method of enzyme immunoassay was used.
Results: At a concentration of 1500 μg / ml and 750 μg / ml. The PI test showed that the placenta extract had no toxic effect on mononuclear cells after 72 hours. But the toxic effect was observed after three days of action of the extract at a concentration of up to 3250 μg / ml, and the toxicity level was 12.0% (p ≤ 0.05). After 2 hours of incubation of cells with placental extract at 1500 μg / ml and 375 μg / ml, a decrease in the ROS level was shown (p ≤ 0.05). Also, the proliferation of T cells was reduced at 1500 μg / ml and 375 μg / ml compared to controls (p ≤ 0.05). It was determined that the increase in IL-4 production with daily exposure depended on the concentration of the placenta extract.
Conclusion: Thus, it was shown that the elements of the horse placental extract have no harmful effects on the mononuclear cells of the human venous peripheral blood. The immunomodulatory effect of placental extract has also been shown.
