Genetic Diversity Assessment of Musa Acuminata (Kluai Hom Thong) and Local Musa. in Central Thailand by Inter Simple Sequence Repeat (ISSR) Fingerprint

Abstract
Musa acuminate (Kluai Hom Thong)is one of an importantcrop in Thailand due to its commercial and nutritional values.A study was conducted to investigate the genetic variability among 6 local banana cultivars in central Thailand using 10 SSR primers. DNA quantity analysis, DNA purification by spectrophotometer,and Inter Simple Sequence Repeats (ISSR)techniques were applied in this study to assess the genetic diversity. The results showed the genomic DNA of Musa Acuminata extraction from three samples were accurated. The sizes of the amplified DNA bands varied from 300 bp to 2,000 bp.The dendogram constructed based on phylogenetic relationship analysis revealed that the highest genetic diversity (0.65)found between Kluai Namwa and Kluai Hom Khieo, Kluai Namwa and Musa Kluai Hom Thong K1, Kluai Namwaand Kluai Hom Thong K2, Kluai Namwa and Kluai Hom Thong K3. The lowest (0.00)was found between Kluai Hom ThongK1 and K2. The UPGMA dendogram has segregated the6 local banana cultivars into two major clusters. Cluster 1 isKluai Hom Khieoand Kluai Hom Thong K1, K2, and K3. Cluster 2 isKluai Namwa and Kluai Hom Khom. The study provedthat Kluai Hom Thong K1, K2, K3 indicated the closer of genetic and obviously different from other cultivars.Moreover,ISSR Fingerprint is universal, quick, easy to apply, highly reproducible, and polymorphous.
