Formulation of Ketoprofen Patch: in Vivo Evaluation on PGE2 Level in Rats

Abstract
Ketoprofen is a lipophilic drug with low bioavailability and short half life therefore should be frequently consumed. The main side effect of this drug as taken orally is GI tract irritation. A special drug delivery system to minimize the side effects this type of drug then become very important issue and transdermal patch is the proposed preparation. The aim of this research was to determine the effect of ketoprofen as anti-inflammatory patch by in vivo studyon PGE2 level in rats induced by carrageenan.Ketoprofen patch were made as three formulas with enhancer variations: F1 (Tween 80 3%); F2 (Tween 80: Oleic acid, 3: 1); and F3 (Tween 80: Oleic acid, 1: 3). Physicochemical evaluation were performed including organoleptic observation, weight variations, thickness, moisture absorption,drug content and drug homogeneity. Anti-inflammatory activity were evaluated in vivo by measuring the levels of PGE2 from carrageenan induced rats using Enzyme Linked Immunosorbant Assay (ELISA). in vivo data showed that ketoprofen from F2 and F3 revealedbetter antiinflammatory activity compared with that from F1. The levels of PGE2 of rats after 3 hours administration of negative control, F1, F2, F3 and marketed product were 1936,84; 1308.71; 803.11; 726.16; and 1162.75 pg/mL respectively. F3 was the best formula as antiinflammatory patch which had the highest % inhibition on PGE2 formation on rats. Keywords - Ketoprofen, Patch, Anti inflammatory, Enhancer, PGE2.
