Test Various Kinds of Temulawak (Curcuma xanthorrhizaRoxb) Dosage forms as Analgesics in Mice with Writhing Reflex Methods

Abstract
Temulawak(Curcuma xanthorrhizaRoxb) known to have analgesic properties because it contains curcumin compounds. The purpose of this study was to determine the analgesic effects of various dosage forms and doses of Temulawak, namely juice, sugar powder and Temulawakextract, by writhing reflex methods. Temulawak (Curcuma xanthorrhiza Roxb) with various dosage forms given to male mice orally with dose variations to determine the percentage of protection, and compared with Acetosal to know the percented ofeffectiveness. This study used 5 Swiss male rats aged 2-3 months with a weight of 20-40 grams for each group, namely 1. Negative control group (1% Na-CMC), 2. Positive control group (acetosal with a dose of 65 mg / kg bb), 3. Group of Temulawak juice with a dose of 6.25%, 4. Group of Temulawak juice with a dose of 12.5%, 5. Group of Temulawak juice with a concentration of 25%, 6. Group of Temulawak sugar doses of 187.5 mg / Kg bw, 7. Group of Temulawak sugar doses of 375 mg / Kg bw, 8. Group of Temulawak sugar doses of 750 mg / Kg bw, 9. Group of Temulawak extract doses of 70mg / kg bw, 10. Group of Temulawakextract dose of 140 mg / kg bw, 11. Group Temulawak extract dose 280 mg / kg bw. To cause writhing reflex given 1% acetic acid induction at a dose of 10 mL / kg bw intra peritoneal, 5 minutes after being treated with Temulawak, and calculated the amount of writhing reflex every 5 minutes for 1 hour. The results of the study showed that the total writhing reflex of the negative control group was 133.4. The average amount of writhing reflex of the positive control group (Acetosal) is 49, percent protection is 63%. The average amount of Temulawak juice writhing reflexdosesof 6.25%, 12.5%, 25% respectively 59.8, 41.6, 26, protection percent 45%, 55.5% and 68.8%; percent of their effectiveness are 71.4%, 87.8% and 108.9% respectively. The average amount of writhing reflex of Temulawak sugar doses of 187.5 mg / kg bw, 375 mg / kg bw, 750 mg / kg bw in a row 106.4, 90.8, 58.8, percent protection is 20%, 32%, and 56 %; percent of effectiveness is 32%, 50%, 88%. The amount of Temulawak extract writhing reflex is 70 mg / kgbw, 140 mg / kgbw and 280 mg / kgbw respectively 82.6, 76.8 and 25.2; percent protection 38.10%, 42.20% and 81.10%; percent of effectiveness is 60.2%, 66.7% and 128.1%. Differences in dosage forms and doses have an effect on Temulawak analgesic activity. The value of IC50 given Temulawak (Curcuma xanthorrizhaRoxb) in the form of juice, sugar powder, and extracts respectively 9.57%, 700 mg / kg bw, and 141, 25 mg / kg bw.
